Classification of Heptatitis C Virus by RT-PCR using Univeral Primers and Sequencing

Abstract

|
Hepatitis C Virus (HCV) in Korea has been classified by RT-PCR and by sequencing 222 by of NS5 region. Among 10 samples whose HCV types were identified by RT-PCR, we found that 2 samples showed different types from sequencing data. These data
suggested
that RT-PCR is not accurate for classfying HCV types. In order to classify HCV types precisely, at least a part of HCV genome should be sequenced. We also found that several different sizes of PCR products were detected from same serum when we
subcloned
PCR products. It may be due to the mispairing of PCR primers to different positions from original sites. It this study, universal primers were designed and used to clone any HCV types.
